GBS (Genotyping By Sequencing)

Genomic DNA of each enset landraces was double-digested with the restriction enzymes, NspI and BfuCI/Sau3AI for prepare the ddRAD-seq libraries. Afterwards two single-strand oligos were ligated to the complementary 3’ and 5’ overhangs of digested sides. The oligo in 3’ overhang sides tagged with a sample-specific unique barcode sequence for sample identification.
